Proteins begin protein synthesis in prokaryotes in the cytosol that has 50S and 30S subunits. If proteins have transmembrane domains, they will contain a signal sequence that will be recognized by the signal recognition particle (SRP), which binds to the sequence and pauses translation. The complex is then taken to the endoplasmic reticulum (ER) where it binds to the translocon and the ribosome is placed on the rough ER. The ribosome restarts translation and the polypeptide is cotranslationally imported into the ER. Modifications to the protein by glycosylation begin in the ER using the oligosaccharyltransferase molecule, which holds onto a block of sugars required for addition onto an asparagine amino acid in the protein sequence. Additional modifications occur in the ERGIC and throughout the Golgi complex. Coated vesicles bud out of the trans-Golgi complex and are transported to their correct molecular address with the help of the appropriate GTPases. The fusion of the vesicle and target membrane occurs at the appropriate destination with the help of proteins that allow the vesicle and target membrane to get close together.
