You are interested in studying a new protein that you have identified called Biggie2. You suspect that Biggie2 may preferentially localize to peroxisomes (and not to other cellular compartments). You would like to use a form of microscopy that allows you to determine the localization (compartment) of Biggie2 in the cell to confirm this idea. Answer both (a) and (b) below. (a) Describe a form of light microscopy (brightfield, fluorescence, etc.) together with sample preparation (e.g. dye labeling) that we discussed that you could use to detect the localization of Biggie2 within cells. Be sure to consider that you are asked to identify the localization of Biggie2, a specific protein, and not proteins in general (not all forms of light microscopy will be useful here). You should include in your answer (i) the name of the method for microscopy and method for labeling of Biggie2, (ii) how these methods allow the detection/labeling of a specific protein and how you would use these methods here, and (iii) what you may expect to see once you use it to determine the localization in the cell of Biggie2. (b) After doing some research, you learn that peroxisomes are often positioned very close to lysosomes. In fact, peroxisomes and lysosomes are ~100 nm apart! You want to make sure that you can tell if Biggie2 is on lysosomes or peroxisomes. (i) Describe the property of light that could prevent you from using the method in your answer in (a) from allowing you to tell if Biggie2 is on peroxisomes or lysosomes. In addition, (ii) describe in detail the method that we discussed (any form of microscopy, light or otherwise) that would allow you to tell if Biggie2 localizes to peroxisomes or lysosomes, which are ~100 nm away from each other (include in your answer how this method works and why it is able to resolve these objects that are 100 nm apart).
