Texts: 1. Phylum Zygomycota Review the life cycle of a typical zygomycete (Rhizopus) in your lecture materials. Examine an intact culture of Gilbertella or Rhizopus from a Petri dish. The dark line in the middle of the plate has formed where mycelia of opposite mating types (the "+" and "-" strains) come together. Looking through, they don't interact with each other. The asexual sporangia are borne on sporangiophore hyphae. Gently scrape small pieces of mycelium containing some sporangia and zygosporangia from a culture plate and prepare a wet mount for examination under the compound microscope. The mycelium is composed of distinct kinds of haploid hyphae: some of these grow through the substrate (rhizoids); others are elongated parallel to the substrate (stolons). Study a portion of the mycelium under high magnification. Note that the hyphae are non-septate (aseptate), that is, they lack cross walls. Thus, the mycelium is multinucleate or coenocytic. In the wet mount, some or most of the sporangia will be broken, exposing the columella (which is the inflated tip of the sporangiophore hypha) and liberating numerous spores (sporangiospores). These spores germinate to produce new mycelia, as a means of asexual reproduction. What are the functions of the rhizoids, stolons, and sporangiophores? How can you tell the difference between an asexual sporangium and a zygosporangium? What is the ploidy of nuclei in the zygosporangium? Where else have you seen structures designated as sporangiophores and how were they different from the sporangiophores of zygomycetes?
