Trypsin is protein digesting enzyme present in the intestine of animals. Though it cannot digest casein (a milk protein), in predator animals drinking the blood of their prey, trypsin hydrolyzes fibrinogen of blood into fibrin, leading to blood coagulation thus help in blood digestion. It also activates other pancreatic proteases.

A. Describe how the following agars work: MacConkey, Blood, Phenylethyl alcohol, Mannitol salt. (DETAILED RESPONSE) MacConkey agar is a selective and differential medium used for the isolation and differentiation of Gram-negative bacteria, particularly members of the Enterobacteriaceae family. It contains bile salts and crystal violet, which inhibit the growth of Gram-positive bacteria and allow the growth of Gram-negative bacteria. MacConkey agar also contains lactose as a fermentable carbohydrate. Bacteria that can ferment lactose produce acid, which lowers the pH of the agar and causes the colonies to turn pink or red. Non-lactose fermenting bacteria appear as colorless colonies. Blood agar is a general-purpose medium used for the cultivation of a wide range of microorganisms, including bacteria, fungi, and some viruses. It is made by adding sterile sheep or horse blood to a nutrient agar base. Blood agar supports the growth of a variety of bacteria and allows for the detection of hemolytic activity. Hemolysis refers to the breakdown of red blood cells, and different types of hemolysis can be observed on blood agar. Alpha-hemolysis results in a greenish discoloration around the colonies, beta-hemolysis causes complete clearing of the agar around the colonies, and gamma-hemolysis shows no change in the agar. Phenylethyl alcohol agar is a selective medium used for the isolation of Gram-positive bacteria, particularly staphylococci. It contains phenylethyl alcohol, which inhibits the growth of Gram-negative bacteria. Phenylethyl alcohol agar also supports the growth of a variety of Gram-positive bacteria, including staphylococci. Staphylococci appear as small, round, opaque colonies on this agar. Mannitol salt agar is a selective and differential medium used for the isolation and identification of staphylococci, particularly Staphylococcus aureus. It contains a high concentration of salt, which inhibits the growth of most bacteria except for staphylococci. Mannitol salt agar also contains mannitol as a fermentable carbohydrate. Staphylococcus aureus can ferment mannitol, producing acid and causing the agar to turn yellow. Other staphylococci that cannot ferment mannitol appear as pink or red colonies on this agar.
